Analyzers

LSR II

  • 4 lasers (488 nm, 407 nm, 633 nm, 355 nm), 13 fluorescent channels
  • Digital acquisition using DiVa software, automatic offline compensation
  • Uses fiber optics and Octagon/Trigon detector arrays to increase sensitivity
  • Detector configuration

Sorters

Aria IIµ

  • 3 lasers (488 nm, 407 nm, 633 nm), 8 fluorescent channels
  • Digital acquisition using DiVa software, automatic offline compensation
  • Uses fiber optics and Octagon/Trigon detector arrays to increase sensitivity
  • Cuvette-based flow cell, more sensitive than jet-in-air cytometer
  • Up to 4-way sorting, single cell sorting into 96 well plates
  • Nozzle size and sorting pressure can be changed for different types of cells. Available nozzles: 70 µm, 85 µm, 100 µm.  Larger nozzle size results in lower sheath pressure, and the cells are less stressed. However, the sort is slower and collected cells are in greater volume of PBS.

Influx with HEPA-filtered enclosure

  • 4 lasers (488 nm, 407 nm, 633 nm, 561 nm), 14 fluorescent channels
  • Analog to digital acquisition
  • 561 nm laser excites PE and PE tandem dyes better than 488 nm laser.  It also excites red fluorescent proteins, such as mCherry, DsRed.
  • Forward Scatter detector is Photo Multiplier Tube (PMT) instead of Diode, more sensitive for particles < 1 µm
  • Jet-in-air cytometer
  • Up to 4-way sorting, single cell sorting into 96 well plates
  • Nozzles: 70 µm, 100 µm, 140 µm, 200 µm.
  • BSL-2 sorting including live human cells

  • Mac Pro computer with Flowjo and CellQuest Pro software
  • CD/DVD writer
